Anyone on the Ascend Plus trial?

In the run in phase, the first 28 days of 3mg run-in was easy, the 2nd 28 days of the 7mg run-in gave me a couple of days of nausea but the 14 mg is causing me to feel dreadfully nauseous most mornings. Positive note is losing 12 lbs but have lost all interest in food, especially in the morning and feeling very tired. Pretty sure I’m on the real drug due to the side effects…it’s difficult but will persevere and am hoping these side effects will pass and I am working on ways to minimilise the nausea as my weight is the issue and this is a real breakthrough for me and hopefully others.

How long have you been on it? I’m on the second bottle of 14mg and I’m not feeling as bad now. But still not got a very good appetite. Sometimes I might think I can tackle a reasonable sized meal but halfway through it suddenly feel full.
Lost interest in a lot of things though.
Nearly at the end of first bottle if 14mg randomised. Definitely semaglutide as the side effects are not nice. Glucose levels v steady as is weight loss. Sticking with it and hope the nausea goes ..

Anyone feel really fatigued too on the trial? 3 weeks into 14mg or placebo and im so tired. Wonder if its down to lack of food?

Same issue here. By mid afternoon, I am so tired and have fallen asleep. I think, for me, it is because I don’t feel like eating because of the nausea so it becomes a vicious circle. I’m now working on eating lots of tiny bits of food throughout the day and using peppermint and ginger to help with the other issues. It’s just trying to get the balance right.

I am a week into my 7mg tablets and am experiencing what feels like vertigo, swimming in head. Has anyone had this and has it eased up ?

Hi Martin, I found in the first week of 7mg that I felt generally lethargic and nauseous but I put it down to a drop in food intake as you just don’t feel like eating. The same goes for the 14mg. No interest in food = no energy. Lost 12lb so far so for me, the benefit outweighs the negatives. It makes you re-evaluate your eating habits. No more big meals.

Do most of you who think you’re taking the ‘proper’ drug vs. placebo in the randomisation period have the bitter taste when taking the tablet? My new randomisation tablets are tasteless and too early to tell if it is placebo, but seems a too obvious difference to use in a drug trial surely

Hi,
I think I am on placebo as randomised tablets are tasteless, like you the run in tablets were bitter, blood sugars have gone up a bit too, but I have been on two holidays since being on randomised.
During run in pills I had no side effects at all, only lost 3lb (which is good as do not have the weight to lose)
Randomised pills no side effects, maintaining new weight.
Having HbA1c in August so will see what that comes in at.

It seemed pretty obvious to me I’m on the Placebo. I went from symptoms to no symptoms.

Well, I started in March 2024 and I am now on my first 14mg bottle. I have so far lost more than a stone in weight. It hasn’t been easy as I had very bad nausea, tummy issues and hypos. The side effects are not that bad now .

I will continue for now as I was aware it was 50% chance I would be on placebo. It am disappointed as run in pills made such a difference to my glucose levels and I had 0 side effects.
I now know there is something out there that works for me. I just hope that in the near future this medication becomes available to diabetic people, without being on a trial or over being overweight.
I just feel five years of taking a placebo is a waste of money after 6 months they could tell us stop the pills and still keep us on the trial list.

Hi, oral semaglutide is available on NHS (UK) However, if you’re prescribed this medication, You have to withdraw from the Ascend Plus clinical trial.

I must be on the real thing, the taste is bitter like the run in. I am on my 2nd bottle of 14mg.,the sides effects are getting worse. First bottle of 14mg, I suffer with very bad nausea all day, gone off of food and could only stomach Cheese and crackers once a day. Now that has eased unless a certain food I smell makes me Nauseous.
Second 14mg bottle, tummy issues, painful trapped wind and diarrhea and still can only manage Cheese and Crackers, any thing else I suffer badly the next day. I drink Peppermint and Ginger Tea, take over the counter anti acid for tummy issues, but it really easing.
I phone the GP to gain a prescription for something stronger than over the counter, gave the symptoms and mentioned it maybe down to the side effects of the drugs. GP had no idea and unable to prescribed anything to help, as the drug was prescribed via Oxford.

Any one suffering the side effect on 2nd bottle plus?? Have you continued? Any suggestion what I can eat to prevent the side effects???

OMG, it seems like you are describing my symptoms. I have had all the symptoms you have .
I can only eat soft foods to avoid stomach pain. Last week I had endoscopy and nothing is wrong with my tummy.
I am on my second bottle and the symptoms are getting better especially my appetite is improving. I had the lowest Hab1c ever-4.9!
I have dropped a whole dress size which I find too much as I wasn’t overweight prior to joining Ascend Plus.
Hang in there, I strongly believe your body will eventually tolerate the symptoms

My recent HbA1C was 39… thanks to the trial. No contact from GP though .. can I come off the metformin if im on what i believe to be the actual semaglutide 14mg now..

No you cannot get off your regular meds as the study is not about HbA1c. The study is about the Heart Health of people with T2 diabetes. You can get more information from the paperworiginal that was sent to us or visit the Ascend Plus website.
