If I have taken Insulin once,as prescribed by the doc,do I have to keep continuing the usage regularly?

Not much information to go on. I can't see a reason why a Dr would prescribe insulin just to take only the once, therefore I would suggest you speak with your Dr about future usage.

Yes not much to go on. But if you are Type 2 diabetic and you improve your blood sugar management, then if your doctor agrees, you may be able to come off it. If you are Type 1 diabetic, then sorry, probably not, barring some new medical breakthrough.
