It's advisable to take metformin with food because it can help with the unfortunate gastric side effects that some people can suffer when taking it.
If you are fasting for 12 hrs you can take your vitamins and metformin when you do eat sometime in the other 12 hrs that you are not fasting

I agree with @catinahat that Metformin should be taken with food to protect your stomach. I take Metformin twice a day, if your dr or nurse says twice a day is ok for you (some people spread it out over 3 doses a day) then you could have one dose at the start of your eating window and the second dose at the end of the 12 hours.

I'm fairly sure that a snall minority can take metformin on an empty stomach without bad effects on the gut. A fair number can't even take extended release on a full stomach.

I can take it on an empty stomach but I would never recommend it to anyone else, it goes against the instructions for taking it for a start. I don’t think there is many of us who can take it like that.
